General description

This product is an extract of E. coli B (ATCC 11303) grown in Kornberg Minimal media at 37°C and taken at 3/4 log growth phase. Total E. coli lipid extract is a chloroform:methanol extract of the respective tissue. This extract is partitioned against deionized water and the chloroform phase is concentrated.

Application

E. coli Extract Total is suitable:
  • for solubilization of His6-tagged wtABCG2 and its steroid-binding element (SBE) mutant variants
  • to prepare E. coli liposomes for liposomal content release assay
  • for the determination of the phospholipid types bound to purified AdiC in DDM (n-dodecyl-β-D-maltopyranoside) and OM (n-octyl-β-D-maltopyranoside)

E. coli extract total has been used for the preparation of liposomes. It has also been used to characterize the ternary complexes obtained after purification of membrane proteins in different detergents.
E. coli Extract Total has been used:
  • in the preparation of small unilamellar vesicles (SUVs)
  • as a reference standard in electrospray ionization tandem mass spectrometry (ESI-MS/MS)
  • in the reconstitution of ATP-binding cassette (ABC) transporter for electron paramagnetic resonance spectroscopy studies

Biochem/physiol Actions

The E.coli lipid extract contains phosphatidylethanolamine, phosphatidylglycerol and cardiolipin. The E. coli lipid extract serves as a standard for profiling phospholipids, aids in the identification and taxonomic grouping of any bacterial sample. It is also used for preparing supported lipid bilayers.
