General description
Myosin-11 (UniProt: P35749; also known as Myosin heavy chain 11, Myosin heavy chain, smooth muscle isoform, SMMHC) is encoded by the MYH11 (also known as KIAA0866) gene (Gene ID: 4629) in human. Myosin-11 (MYH11) is a smooth muscle-specific myosin heavy chain protein encoded by the MYH11 gene located at chromosome 16p13.11. Myosin-11 consists of a motor domain at its N-terminus responsible for ATPase activity and actin binding, along with a coiled-coil tail domain enabling dimerization and cargo transport. The protein plays a critical role in smooth muscle contraction, intracellular transport, and cytoskeletal organization by interacting with actin filaments. Myosin-11 is predominantly expressed in smooth muscle tissues, including vascular and gastrointestinal systems, associated with actin filaments. It is involved in pathways such as cytoskeletal remodeling regulated by Rho GTPases and semaphorin interactions, which influence cellular motility and morphology. Myosin-11 interacts with other proteins, including receptor proteins on organelles, to mediate transport along polarized actin cables. Mutations in MYH11 have been linked to familial thoracic aortic aneurysms and dissections (TAAD), acute myeloid leukemia (AML), as well as megacystis-microcolon-intestinal hypoperistalsis syndrome (MMIHS). Overexpression of MYH11 has also been shown to induce endoplasmic reticulum stress and autophagy, which may contribute to vascular pathology. Application
Quality Control Testing
Evaluated by Western Blotting in rat lung tissue lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Myosin-11 in rat lung tissue lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Myosin-11 in human uterus, human prostate, and mouse lung tissue lysates.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Myosin-11 in human uterus tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound with Myosin-11 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 1L21 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Myosin-11. It targets an epitope within 22 amino acids from the N-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
