General description
Research area: Cell signaling. Osteopontin is initially secreted as a 298 amino acid protein, which is subject to multiple post-translational modifications including glycosylation, phosphorylation, and specific proteolytic cleavages into various smaller molecular weight fragments. Osteopontin is expressed in a wide range of cells and tissues including osteoblasts, various tumor cell lines, extraosseous cells in the inner ear, brain, kidney, deciduum, placenta, and odontoblasts. Recombinant human Osteopontin is a 298 amino acid protein, which, due to glycosylation, migrates at an apparent molecular weight of 60.0-65.0kDa by SDS-PAGE analysis under reducing conditions.
Application
Osteopontin human has been used in in vitro invasion assay to study the effect of endostar on non-small cell lung cancer cell invasion. It has been used in the cell culture of primary human brain vascularsmooth muscle cells. Osteopontin human has also been used to study itseffects on the migration of human fibroblasts.
Biochem/physiol Actions
Osteopontin is a secreted glycoprotein that functions as a ligand to αvβ3 integrin and possibly other receptors. It binds tightly to hydroxyapatite and can act as a structural component of the extracellular mineralized matrix. In addition to its involvement in mineralized matrix formation, Osteopontin can also function as a cytokine that stimulates the release of IFNg (interferon γ) and IL-12 (interleukin 12), while inhibiting the production of IL-10. Osteopontin is involved in inflammation and also regulates immunity against infectious diseases. It also co-stimulates T cell proliferation. Osteopontin is also associated with multiple sclerosis. It participates in inflammation in Crohn′s Disease. Osteopontin plays a crucial role in bone remodelling and suppresses ectopic calcification.
Physical form
Lyophilized from 10 mM Sodium Phosphate, pH 7.0.
Preparation Note
Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.

Osteopontin is an acidic glycoprotein of about 41,500 daltons that has been isolated from rat, human and bovine bone. It is rich in aspartic acid, glutamic acid and serine and contains about 30 monosaccharides, including 10 sialic acids. Several types
