General description
10% Neutral Buffered Formalin is the most used fixative throughout the world for light microscopy. Formalin solution is the commercially obtained solution containing 37% w/w (40% w/v) of formaldehyde in water. It requires a relatively short period of fixation. It is suitable for the long-term storage of tissue. Once properly fixed, the tissue should be able to withstand the subsequent stages of tissue processing or staining.
Application
Formalin solution (neutral buffered, 10%) has been popularly used as a general-purpose histological fixative to fix tissue samples.
- It effectively preserves the cellular and tissue morphology, maintaining the structural integrity required for microscopic examination.
- It is used for routine histological staining, providing clear and detailed images of cellular structures.
Popular general purpose histological fixative.
Biochem/physiol Actions
Formaldehyde is both a non-coagulant and an additive fixative. Formalin fixation is thought to form cross links between the aldehydes and the proteins, creating a gel, thus retaining cellular constituents in their in vivo relationship. Formaldehyde is known to penetrate tissue quickly, but the fixation process may take a long time to form the various cross links with tissue proteins.
Features and Benefits
- 10% NBF is well-suited for large throughput laboratories.
- It requires a relatively short period of fixation.
- It can be used for the long-term storage of tissue.
Packaging
Case containers are polypropylene with white leak-proof lids and are partially prefilled with 10% NBF.
