Application
Bromelain from pineapple stem has been used as a positive control in sodium dodecyl sulfate (SDS) / polyacrylamide gel electrophoresis (PAGE) zymograms. It has also been used as a Azocasein substrate for the determination of proteolytic activity.
Bromelain may be used to inhibit the biosysnthesis of proinflammatory prostaglandins. It may be used to reduce clotting efficiency. Bromelain, from pineapple stem, has been used to make enzymatic hydrolysates of honeybee-collected pollen .
Biochem/physiol Actions
Bromelain is a cysteine endopeptidase with broad specificity for cleavage of proteins. Bromelain may be from a stem or piece of fruit. Product P3169 (pGlu-Phe-Leu-p-nitroanilide) may be used as a chromogenic substrate for the assay of bromelain.
Physical form
Lyophilized powder containing mannitol and potassium phosphate buffer salts
Preparation Note
Chromatographically purified
Chromatographically purified. Dissolves in water at 1 mg/mL concentration to form a clear, colorless solution.
Other Notes
One unit will release 1.0 μmole of p-nitrophenol from N-α-Z-L-lysine p-nitrophenyl ester per min at pH 4.6 at 25 °C.

Stem bromelain was covalently coupled to a thermosensitive polymer of N-isopropylacrylamide (p(NIPAm)) either through the amino groups of the enzyme (randomly coupled) or via the lone oligosaccharide chain (uniformly coupled). The enzyme coupled via the oligosaccharide chain exhibited better access
