Integration of Automated Sampling with LC-MS as a PAT Tool for USP Glycsoylation Monitoring and Control

The MAST^® Autosampling Solution and Agilent 2D-LC/TOF MS was used to enable glycosylation monitoring during perfusion cell culture. A digital lab infrastructure and cloud-based orchestration layer managed communication and control between the units, allowing easy modification of parameters like target galactosylation and sampling frequency. Over a 35-day culture process, targeted galactosylation setpoints were maintained within a ± 2% deadband, ensuring efficient data collection and timely insights.
